Course content
- Instruction for security of work, formation of working groups - Preparation of chloroplast suspension - Emission and excitation spectra of chlorophyll - explanation of the method, meaning of evaluated parameters, measurement of the spectra by spectrofluorometer Hitachi F4500 in room temperature and 77 K using control and stressed samples - Slow fluorescence induction and fluorescence quenching - explanation of the method, meaning of evaluated parameters, measurement of the induction and quenching by fluorometer PAM 2000 using control and stressed samples - Very fast fluorescence induction - explanation of the method, meaning of evaluated parameters, measurement of the induction by fluorometer PEA using control and stressed samples - Light induced changes in emission spectra - explanation of the method, meaning of evaluated parameters, measurement of the spectra at room temperature by spectrophotometer Olis using control and stressed samples - Synchronous detection of very fast fluorescence induction and A820 signal - explanation of the method, meaning of evaluated parameters, synchronous measurement of the induction and A820 signal by fluorometer Dual PAM 100 using control and stressed samples - Very fast detection of P515 signal - explanation of the method, meaning of evaluated parameters, measurement of the P515 signal by fluorometer Dual PAM 100 using control and stressed samples - Fluorescence imaging - explanation of the method, meaning of evaluated parameters, measurement of the imaging by fluorometer FluorCam using control and stressed samples - Fluorescence temperature curve - explanation of the method, meaning of evaluated parameters, measurement of the curve by laboratory-made apparatus using control and stressed samples
