Course content
Identification of amino acids. Determination of the isoelectric point of casein. Detection of the presence of glutamic acid in food flavorings. Qualitative and quantitative analysis of carbohydrates in sweeteners. Determination of proteins by Bradford method in tube and microplate arrangement. Isolation of amine oxidase from pea seedlings and determination of its activity. Activity, substrate specificity and pH optimum of enzymes. Isolation of Ri-plasmid DNA from the bacterial strain Agrobacterium rhizogenes R15834 by alkaline lysis. Preparation of lipid fractions from egg yolk. Separation methods. Gel chromatography. Agarose electrophoresis. Determination of isoenzyme representation of plant peroxidases by discontinuous electrophoresis in polyacrylamide gel under native conditions.

Student performance

CONDITIONS FOR SUBJECT COMPLETION 1. 100% attendance at the seminar. In case of illness or other serious reasons, one exercise can be replaced within another group, only once per semester (exceptional situations will be dealt with individually, e.g. hospital stay). 2. Submitted and recognized protocols from all exercises. A pair of students solving a given task together submit one common protocol in electronic form. The elaborated protocols must be submitted no later than 7 days after the task has been completed! For each protocol duly submitted by the deadline, which is correctly prepared, the student will receive 5 points. If, on the other hand, the protocol or its correction is not submitted by the due deadline (i.e. within 7 days), 5 points will be deducted. If the corrected protocol is returned to the student (not all errors or deficiencies are corrected), 2 points are deducted each time. If it is found that the protocol was copied (e.g. the results will not correspond to the data given in the workbook), the exercise is invalid and the student cannot obtain the credit. Students must also submit the protocol from the last exercise within 7 days of completing the task, in case of need for correction, the protocol can be returned only once, after which the points for uncorrected deficiencies are deducted adequately. All protocols must be recognized no later than 4 weeks from the date of the last exercise. 3. During the exercises, the student's readiness will be tested in the form of a test (5 questions from the theoretical and practical part of the task + 1 numerical example). - A well-prepared student can obtain 6 points from the test. - If the student obtains 2-3 points from the test, the supervisor is obliged to examine the student orally, in case of ignorance of the topic, the student is expelled from the exercises. If the student was not satisfied with the type of questions in the test, but is familiar with the topic, then he continues with the exercise. The number of points from the test remains the same. - A student who gets less than 2 points from the test does not continue in the exercise and must make up for it. Due to unpreparedness, it is possible to repeat the seminars only once per semester. Students who attend a smaller number of exercises due to public holidays, planned practice or excursion, as well as third-year students who finish earlier, will receive the corresponding number of points according to the rules. CREDIT The student must meet all requirements for completing the course (see above) and obtain the necessary number of points for readiness and submitted protocols. 75 - 100 % of points - credit is granted. 50 - 75 % of points - the student will write a credit test (the test must be written with more than 70 % of points for the credit, the credit test can be repeated only once). < 50% of points - the student will not be awarded the credit, he/she must enroll in the course again next year.
