Course content
History of the microscopy. Physical characteristics of the optics in the light microscopy. Basic components of the light microscope: objectives, ocular, light, numeric aperture and magnification. Working rules with microscope; drawing, measurement and calculation of the microscopical objects; image analysis. Advanced microscopic methods: dark field, phase contrast, polarization, interference, Nomarski Differential Interference Contrast (DIC). Fluorescence microscopy: mechanism, basic components of fluorescent microscope, fluorescent dyes, basic fluorescent methods, cytospectroscopy. Principles of the confocal microscopy, basic confocal methods; various way of improvement the microscope image quality. Various types of microscopes: stereomicroscope, inversion microscope, 3D microscopy, STM and ASM microscopes; microscope as the micromanipulator. Preparation of the microscopic slides: native, permanent, smearing, squash and cutting; fixation of the materials, microscopic dyes. Electron microscopy: mechanism of the TEM and SEM, preparing of the electronmicroscopic slides. <br>Excursions: Electronmicroscopes - TEM and SEM (RCPTM - Department of carbon nanostructures, UP Olomouc). Microscopic equipments of the scientific laboratory (ÚEB Olomouc); Confocal microscope (Department of Cell Biology, Centre of the Region Haná, Olomouc).

Learning outcomes
Light and electron microscopy, basic and advanced microscopic methods. Preparation of the microscopic slides and their documentation.
Student is able to (after attending the course): - Describe principles of image appearance in light and electron microscope. - Describe detailed structure of light microscope. - Explain principles of contrast microscopy. - Draw, measure and count in the microscope. - Prepare various types of microscopic slide. - Describe the main types of electron microscope (TEM, SEM), and preparation of EM slides.
