1) Safety guidelines and hygiene in a molecular biology laboratory. Basic conventions, units, and terminology. 2) Isolation of genomic DNA from plant tissues, determination of concentration and purity (spectrophotometry, nucleic acid gel electrophoresis). 3) Avian sex determination - PCR amplification of genomic DNA and electrophoretic analysis. 4) Isolation of immunoglobulins from blood serum, determination of IgG heavy and light chain MW (SDS-PAGE protein electrophoresis). 5) Total RNA isolation from plant tissues using a spin-column kit, determination of concentration and purity (spectrophotometry, gel electrophoresis). 6) Transformation of competent bacteria with plasmid DNA, antibiotic resistance transfer, and blue-white screening of transformed colonies.

| Learning activities and teaching methods |
| Dialogic Lecture (Discussion, Dialog, Brainstorming), Laboratory Work |
| Learning outcomes |
|
To acquire practical skills in core molecular biology techniques - namely the extraction and analysis of nucleic acids (DNA, RNA) and proteins, PCR, and bacterial transformation.
Upon successful completion of the course, the student will be able to: Apply acquired laboratory skills to practical research in molecular genetics. Perform fundamental techniques in molecular biology research. Write a structured experimental report and critically discuss the results. |
