Course content
1) Laboratory safety and hygiene in a biological laboratory. Fundamentals of biological microscopy techniques, structure and correct use of a light microscope, principles of microscopy, types of microscopic specimens, principles of biological drawing and laboratory documentation. 2) Preparation of simple microscopic specimens and staining of biological material. Observation, biological drawing, scientific description, and documentation of observed structures. 3) Microrelief method - preparation of leaf epidermal impressions from selected plant species, comparison of stomatal impressions of the male fern and cock's-foot grass, or other selected plants. Observation of the epidermis and stomatal apparatus and identification of stomatal types. 4) Preparation of transverse sections of needles of different pine species using elder pith. Anatomy of conifer needles, observation and description of anatomical structures, and identification of selected pine species based on anatomical characteristics using an identification key. 5) Preparation of transverse sections of plant material using elder pith. Observation of the anatomy of plant organs, staining of lignified structures with phloroglucinol or safranin, and measurement and counting of microscopic structures. 6) Practical training and assessment of basic biological microscopy techniques. Independent preparation of microscopic specimens, microscopic observation, biological drawing, scientific description, and interpretation of results. Preparation of laboratory documentation and reflection on practical skills.

Learning outcomes
The aim of the course is to develop basic laboratory skills and biological microscopy techniques. Students will learn to work safely in a biological laboratory, use a light microscope correctly, prepare and examine microscopic specimens, document and scientifically interpret observed biological objects, and present the results of practical work in the form of laboratory reports. Emphasis is also placed on independent laboratory work, adherence to proper laboratory procedures, and reflection on students' own practical skills.
Student should be able to (after attending the course): - follow health and safety and hygiene principles in a biological laboratory; - explain the principles of basic biological microscopy techniques and laboratory procedures; - use a light microscope correctly and independently; - prepare microscopic specimens using selected preparation and staining techniques; - observe, draw, accurately describe, and interpret biological specimens; - record laboratory procedures and results and present them in the form of a laboratory report; - analyse the procedures used and evaluate the results of observations; - reflect on their own practical skills, identify potential errors, and use feedback to improve their subsequent laboratory work.

- regular theoretical preparation for assigned practical tasks and demonstration of knowledge of their principles and procedures; - active participation in practical classes and mastery of basic microscopy techniques; independent preparation of microscopic specimens, microscopic observation, biological drawing, scientific description, and interpretation of results; - continuous preparation of laboratory reports, including their revision and completion based on feedback from the instructor; - compliance with safety and hygiene principles in the biological laboratory; continuous self-assessment of practical skills using a self-assessment form; - successful practical demonstration of basic microscopy skills through the independent preparation of a microscopic specimen, its microscopic observation, scientific description, and interpretation of results according to predefined criteria. - completion of a seminar paper according to the assignment requirements and correction of any errors and deficiencies based on feedback from the instructor.
