Course content
Theoretical part Fundamental principles of solid-phase organic synthesis. Definition of the types and properties of resins and linkers, and their application according to the type of the synthesized molecule and reaction conditions. Mechanisms of individual reaction steps, including the cleavage of (intermediates) products from the resin. Experimental part: - Immobilization of the first reagent onto the solid phase according to the selected type of resin and linker and determination of resin loading. - Design of the reaction sequence and synthesis of target motifs in line with current trends in organic chemistry, e.g., linear or cyclic peptides or heterocyclic derivatives. - Utilization of Fmoc-protected amino acids in solid-phase synthesis and evaluation of various coupling reagents for amide bond formation. - Modification of functional groups on the solid phase or in solution. - Application of protecting groups in solid-phase organic synthesis, including orthogonal protection/deprotection strategies. - Application of various conditions for cleavage from the solid support to obtain structurally different products. - Introduction to UPLC/MS analysis of (intermediates) products, purification methods, and determination of synthetic yield.

Learning outcomes
To familiarize students with the fundamental principles and practical skills of solid-phase organic synthesis, with emphasis on the design of an appropriate synthetic strategy for selected target molecules, including the choice of a suitable resin and linker, protecting groups, and efficient cleavage from the solid support. The primary focus of the course is the practical part, in which students perform assigned tasks within the framework of laboratory exercises.
