Course content
|
1) Introduction, organization of exercises, safety in the laboratory. 2) Preparation of cultivation media, sterilization, disinfection. Microorganisms in the environment, morphology of microorganisms on solid culture media, microbial growth in liquid media. Inoculation and isolation of microorganisms (aseptic technique, streak plate technique, serial dilution technique). 3) Determination of number of microorganisms (culture and microscopic techniques). 4) Influence of different factors on microbial growth. Bacterial resistance against antibiotics caused by plasmid DNA. 5) Physical and chemical methods of microbial growth control (effect of UV radiation, disinfectants, antibiotic susceptibility testing by diffusion method, identification of bacteria degrading ß-lactam antibiotics and antibiotic susceptibility testing by dilution method). 6) Microscopic examination of microorganisms, use of staining techniques. 7) Principles of bacterial identification: biochemical identification tests. Rapid identification of Enterobacteriaceae and identification of staphylococci at the genus level. 8) Microorganisms in soil: demonstration and isolation of selected soil microorganisms. Bacteria in water: basic microbiological analysis of drinking water. Microorganisms in food: bacteria in yoghurt, analysis of meat sample. Cultivation of anaerobes from yogurt, meat and water. 9) Eukaryotic microorganisms: characteristics of yeast and fungi.

Learning outcomes
|
The goal of this course is to teach students basic microbiological techniques and to show them the importance of microorganisms in nature and in daily life. Principles of microbiology are demonstrated using practical examples.
Ability to apply fundamental microbiological procedures and methods .

Prerequisites
|
For 2nd year students of the Biochemistry study program and the Biotechnology and Genetic Engineering study program.

Assessment methods and criteria
|
Student performance, Systematic Observation of Student
Full participation at the practicals is required (maximum two absences may be tolerated for serious reasons and the experiments may be conducted in a different class as decided by the teacher); all experimental tasks must be completed, reports submited each task and prerequisite points obtained for the reports and in written tests.
