Course content
Techniques of nucleic acid isolation and purification, determination of their concentration and quality. Nucleic acid gel electrophoresis. Enzymes used in molecular biology (polymerases, nucleases, ligases, phosphatases, kinases, methylases). Hybridization of nucleic acids, hybridization probes. Polymerase chain reaction (PCR, RT-PCR), quantitative PCR (qPCR, qRT-PCR), their chemistry and evaluation. Sanger and NGS sequencing. DNA cloning, cloning and expression vectors, in vitro transcription, in vivo translation. Targeted mutagenesis. Determination of protein-protein, nucleic acid-protein interactions. Molecular methods of genetic variability of organisms and their population analysis (RFLP, AFLP, RAPD, SNP detection, SSCP, microsatellite DNA).

Learning outcomes
Introducing students into the theoretical basis, techniques, evaluation and application of modern methods of molecular biology and genetics. Emphasis is placed on the ability of the students to choose appropriate techniques in realization of experiments.
Student is able to (after attending the course): - Explain the principles of molecular and genetic methods and their application. - Describe methods DNA isolation and purifications, probe labelling, hybridisation techniques, construction of recombinant DNA, PCR and its variants, methods directed mutagenesis, methods of evaluation the variability a genetic diversity of populations and organisms. - Propose conception and methodological approach for molecular and genetic experiments.

Assessment methods and criteria
Oral exam

Oral exam and evaluation of experimental data in extent of the lectures, 2 randomly selected questions, practical evaluation of experimental data, work with information sources.
