Course content
|
Izolation of bacterial recombinant plazmid. Polymerase chain reaction (PCR) - aster yellows phytoplasma detection . Random fragment lenght polymorphism (RFLP) and gel electrophoresis in agarose gel. Analysis of microsatelite DNA locus polymorphisms. PCR In situ (PRINS) - physical localization of repetitive sequences on chromosomes. Fluorescence in situ hybridization (FISH) - physical localization of chromosome repetitive sequences.

Learning outcomes
|
Student should be able to (after attending the course): - isolate recombinant plasmids using ´spin´ tubes. - run a Polymerase Chain Reaction. - perform RFLP analysis of the PCR product. - perform in situ PCR on isolated chromosomes. - detect polymorphism of microsatellite loci. - analyse results and write a report (about aims, methods and results of the experiments).
