Course content |
The subject focuses on the principles, history and modern applications of light microscopy (including fluorescence and confocal m.) and other imaging techniques (SEM, TEM, atomic force microscopy, electron tomography), on the basic principles of specimen preparation for various microscopic methods, including correlative microscopy, on editing of microphotographs for publication purposes, on the mathematical methods for digital data processing, image analysis (histograms editing, dimension measurement, counting objects, 3D models). 1. Introduction, history, view, overview and comparison of direct and indirect methods of microscopy 2. Light microscopy and its modifications (phase contrast , polarized light, darkfield , Nomarski DIC) lenses 3. Fluorescence microscopy - principle of fluorescence, fluorochromes , transmission and epifluorescence microscopes, deconvolution 4. The preparation of biological samples for fluorescence microscopy - histochemical and immunohistochemical localization, fusion proteins 5. Confocal microscopy - spinning disc CM, laser scanning CM, multiphoton CM 6. Electron microscopy - history, principles, vacuum 7. SEM - scanning electron microscopy 8. TEM - transmission electron microscopy 9. Scanning tunneling microscopy, atomic force microscopy 10. Other imaging methods and their combinations (correlation microscopy, near-field optical microscopy, magnetic resonance imaging) 11. Acquaintance with current trends in microscopy - invited speakers 12. Acquaintance with current trends in microscopy - short presentation of students on a selected topic
